技術(shù)原理

CellTiter-Glo? Luminescent Cell Viability Assay (CellTiter-Glo?發(fā)光法細(xì)胞活力檢測(cè)試劑盒) 是通過對(duì)ATP 進(jìn)行定量測(cè)定來檢測(cè)培養(yǎng)物中活細(xì)胞數(shù)目的一種均質(zhì)快速檢測(cè)方法。其主要原理是利用螢光素酶在與甲蟲螢光素底物進(jìn)行發(fā)光反應(yīng)時(shí)需要ATP，而ATP的量與發(fā)光強(qiáng)度呈線性關(guān)系。
試劑盒中包括緩沖液及凍干底物，檢測(cè)前將緩沖液與檢測(cè)底物完全混合形成 CellTiter-Glo? 試劑，檢測(cè)步驟就是將單一試劑(CellTiter-Glo? 試劑) 直接加入含有血清的培養(yǎng)細(xì)胞中，無需洗滌細(xì)胞、去除培養(yǎng)基或進(jìn)行多步加樣操作。在384孔板上，加入試劑并混合后，10分鐘內(nèi)，該系統(tǒng)可檢測(cè)到的每個(gè)孔內(nèi)的細(xì)胞數(shù)最低為15個(gè)。

CellTiter-Glo? Luminescent Cell Viability Assay 在很寬的細(xì)胞數(shù)范圍內(nèi)都呈現(xiàn)線性
左圖 . 細(xì)胞數(shù)與發(fā)光信號(hào)相關(guān)。 在 96 孔板中以 RPMI 1640 + 10% FBS 進(jìn)行 2 倍梯度稀釋 Jurkat 細(xì)胞。再加入 CellTiterGlo? Reagent，孵育 10 min 后檢測(cè)?？梢娫?0-50000 個(gè)細(xì)胞/ 孔 (96 孔板 ) 的范圍內(nèi)與光信號(hào)呈線性相關(guān) (r2 = 0.99) 。
